Chk2 is a protein encoded by the CHEK2 gene which is approximately 60,9 kDa. Chk2 is localised to the nucleus. It is involved in CDK-mediated phosphorylation and removal of Cdc6, regulation of TP53 activity and apoptotic pathways. It is a serine/threonine-protein kinase which is required for checkpoint-mediated cell cycle arrest, activation of DNA repair and apoptosis in response to the presence of DNA double-strand breaks. It contains a forkhead-associated protein interaction domain essential for activation in response to DNA damage and is rapidly phosphorylated in response to replication blocks and DNA damage. Chk2 is highly expressed in the testis, spleen, colon and peripheral blood leukocytes. Mutations in the CHEK2 gene may result in Li-Fraumeni syndrome. STJ90225 was affinity-purified from rabbit antiserum by affinity-chromatography using epitope-specific immunogen. This polyclonal antibody detects endogenous levels of Chk2 protein only when phosphorylated at T68. This primary antibody specifically binds to endogenous Chk2 protein which only binds about T68 when T68 is phosphorylated.
